Takeda Advances in Dermatology with Promising New TYK2 Inhibitor: An Insight into Zasocitinib

Dr. Christopher Bunick, MD, PhD, also provided additional context on the significance of these findings.
Zasocitinib Design and Development
Developed by a team of chemists at Nimbus Therapeutics in collaboration with Schrödinger for AI-assisted design, zasocitinib demonstrates exceptional precision in targeting the TYK2 enzyme, a critical component in treating immune-mediated inflammatory diseases. The AI integration enabled the efficient creation and screening of numerous compounds, ensuring the best possible fit for the targeted enzyme.
“Zasocitinib was designed through a collaborative effort employing advanced AI tools to enhance the accuracy and efficiency of drug development,” said Dr. Heap. This meticulous design process has resulted in a highly selective inhibitor, with the drug binding to the TYK2 site 1.7 million times more than the JAK1 site, vastly outstripping the selectivity of existing inhibitors.
Pre-clinical and Clinical Data
In laboratory settings, zasocitinib exhibited exceptional binding affinity and selectivity. Compared to deucravacitinib, the currently available TYK2 inhibitor, zasocitinib demonstrated a 19,000-fold increase in selectivity for TYK2 over JAK1.
The in vitro studies were validated through ex vivo experiments, where zasocitinib maintained 91% inhibition of the TYK2 pathway over 24 hours, a marked improvement over the 24% inhibition observed with deucravacitinib. These findings suggest that zasocitinib could offer significant therapeutic benefits with continuous and effective inhibition, setting it apart from both current TYK2 and JAK inhibitors.
Future Research and Trials
Takeda, a company with 243 years of pharmaceutical heritage, is now making significant inroads into dermatology. "The next step is to proceed with phase 3 trials. We're looking to see what the drug can achieve in a larger, more prolonged setting," said Dr. Heap. These phase 3 trials are ongoing, particularly in patients with psoriasis, aiming to validate the promising phase 2 results where approximately one-third of patients achieved clear skin at the 12-week mark.
Additionally, Takeda is exploring the efficacy of zasocitinib in treating psoriatic arthritis, with phase 3 trials set to commence in the coming year. Preliminary results from phase 2 studies will be presented at the upcoming European dermatology meeting.
The investigational drug is also being tested for its potential in treating inflammatory bowel diseases, including Crohn’s disease and ulcerative colitis, with ongoing phase 2 studies recruiting participants.
