In a notable development within the biotechnology sector, Archon Biosciences has announced its emergence from a stealth mode, bolstered by $20 million in seed financing. This new phase for the company is poised to explore further depths of therapeutic innovation through its pioneering technology involving Antibody Cages (AbCs). The company is focusing on utilising AbCs—an entirely new class of biologic medicines characterised by novel geometric configurations of antibodies—to target therapeutic challenges that have been beyond the reach of existing modalities.

AbCs represent a revolutionary divergence from traditional biologic medicines, presenting a platform where natural antibody properties are preserved while additional characteristics can be meticulously adjusted. This innovative structure allows for modifications in the way these biologics distribute and retain within the body, as well as the manner in which they engage with their targets.

The initial research into AbC technology was detailed in a 2021 publication in Science, under the title, "Designed proteins assemble antibodies into modular nanocages.” Archon now seeks to harness the potential of its proprietary protein design platform through its AI-driven approach, making significant strides in biologics by addressing medically intractable targets.

Dr. James Lazarovits, co-founder and Chief Executive Officer of Archon, emphasises the potential impact of AbC technology. According to Lazarovits, it aims to tackle pivotal disease targets effectively, creating improved treatment options for patients. He cites the backing of investors and the company’s in-house capabilities for rapid manufacturing and testing as crucial components in their quest to redefine biologic development.

The platform developed by Archon is rooted in computational protein design innovations that earned the 2024 Nobel Prize in Chemistry. This accolade was shared by Dr. David Baker, a professor of biochemistry at the University of Washington and the Howard Hughes Medical Institute investigator. Baker co-invented the Archon platform with Lazarovits and George Ueda, PhD, who now serves as the company's Chief Technology Officer.

The generative protein design approach employed by Archon involves the creation and manufacture of new molecular entities for preclinical evaluation. This process ingeniously merges off-the-shelf antibodies with computationally designed binding proteins, assembling them into highly precise nanostructures. These meticulously crafted geometries result in AbCs behaving and interacting with biological targets in distinct ways, offering enhanced control over properties such as target engagement and biodistribution.

Manufacturing these AbCs doesn't require altering the traditional antibody production process, allowing for a massive parallelisation that is readily automated. This ensures the retention of known binding affinities and specificities while also opening the door to new geometric designs and applications.

Leading the financing round for Archon's venture is Madrona Ventures, with additional support coming from prominent investors such as DUMAC, Sahsen Ventures, WRF Capital, Pack Ventures, Alexandria Venture Investments, and Cornucopian Capital. This financial backing positions Archon to advance its pioneering work in computational protein design, fostering a new era of tailored biologic therapeutics.
